fa ارتباط بین پلی مورفیسم VNTR ژن IL-4 با دیابت نوع دو Association between IL-4 Gene VNTR Polymorphism and Type 2 Diabetes تخصصي Special پژوهشي Original مقدمه و هدف: یکی از بیماری های رایج که در تمام کشورها افراد بسیار زیادی به آن مبتلا هستند، دیابت می باشد. از جمله عوامل مهمی که در ایجاد و گسترش این بیماری نقش دارد، التهاب می باشد. اینترلوکین 4 به عنوان یکی از سایتوکاین های ضد التهابی بوده که می تواند سبب کاهش التهاب گردد. هدف از انجام این مطالعه بررسی ارتباط  پلی مورفیسم VNTR ژن IL-4 در افراد مبتلا به دیابت نوع 2 در شهرستان گناباد کشور ایران می باشد. مواد و روش‌ها: این پژوهش  یک مطالعه مورد شاهد است. 100 نفر از افرادی که به بیمارستان علامه بهلول  شهرستان گناباد  مراجعه کرده بودند، در دو گروه مورد (مبتلا به دیابت نوع دو) و شاهد (سالم) قرار گرفتند. انتخاب افراد به صورت تصادفی انجام شد و در صورت داشتن معیارهای ورود، پس از اخذ رضایت آگاهانه، وارد مطالعه شدند. از هر فرد 2 میلی لیتر خون با استفاده از لوله حاوی EDTA جمع آوری شد و استخراج DNA با استفاده از روش salting out صورت گرفت. برای بررسی پلی مورفیسم، ابتدا قطعه مورد نظر با استفاده از پرایمرهای مناسب تکثیر داده شد و سپس با استفاده از الکتروفورز شناسایی شد. یافته‌ها: میانگین سنی واحدهای پژوهش 7/30±61/98سال بوده و نیمی از ایشان مرد بودند. نتایج بررسی پلی مورفیسم نشان دهنده عدم ارتباط معنی دار بین پلی مورفیسم VNTR ژن IL-4 و ابتلا به دیابت نوع دو بود (05/0P >). بحث و نتیجه‌گیری: در جمعیت مورد مطالعه ما، ارتباط معنی داری بین پلی مورفیسم VNTR ژن IL-4 با دیابت نوع دو وجود نداشت.   Abstract: Background and Objective: Diabetes mellitus is a prevalent disease affecting a large population worldwide. Inflammation plays a crucial role in the development and progression of this disease. Interleukin-4 (IL-4) is an anti-inflammatory cytokine that can reduce inflammation. The aim of this study was to investigate the association between IL-4 gene VNTR polymorphism and type 2 diabetes in Gonabad city, Iran. Methods: This study was a case-control study. One hundred individuals who referred to Alameh Behlol Hospital Gonabad city of Iran were included in two groups: case (patients with type 2 diabetes) and control (healthy individuals). The participants were randomly selected and entered the study after obtaining informed consent and meeting the inclusion criteria. Two milliliters of blood were collected from each participant using EDTA-containing tubes, and DNA was extracted using the salting-out method. To investigate the VNTR polymorphism, the target fragment was first amplified using appropriate primers and then identified using electrophoresis. Results: The mean age of the study participants was 61.98 ± 7.30 years, and half of them were male. The results of the VNTR polymorphism analysis demonstrated no significant association between IL-4 gene VNTR polymorphism and type 2 diabetes (p > 0.05).
